چکیده

THE ENVIRONMENT AND THE LUNG 39$ achieved by prior culture for five days in DMEM + 0.5% FCS) and fibroblast cell counts determined after one, two, and four days. DMEM + 0.5% FCS ± asbestos incubated for four hours as controls. Supernatants from unexposed PAM had no influence on fibroblast replication rate. In contrast, supernatants from PAM exposed to asbestos significantly increased the rate of fibroblasts doubling; the doubling time decreasing five-fold from a control of 310±21 hours to 54±4 hours, P<.001. The stimulation of fibroblast replication was readily apparent by 48 hours with 68±2 X 10 cells per plate in the group treated with asbestos exposed PAM supematants compared to only 43±3 X 108 cells per plate in the control groups. This study demonstrates that asbestos-treated PAM release factor(s) promoting human lung fibroblast replication, and suggests a possible role for PAM in the formation of the fibroblast clusters observed in the early lesions of asbestosis.

Interaction of asbestos with alveolar cells

A number of electron microphotographs are presented showing the various aspects of phagocytosis of fibers in lung tissue. The fibers were rapidly phagocytosed by alveolar macrophages, by polymorphonuclear leucocytes, and less frequently by alveolar epithelial cells. They were also found in the cytoplasm of the alveolar stromal macrophages. In the late stage of the disease, macrophages containin...

Safety Evaluation of Rw after Nasal Inhalation in Rats

Asbestos is reported to cause pulmonary fibrosis, and its use has been banned. We examined the biopersistence and histopathological effect of rock wool (RW) fibers in rat lungs by a nose-only inhalation exposure study. Twenty-four rats were exposed to RW fibers for 6 h daily for 5 consecutive days. Six rats each were sacrificed shortly and 1, 2, and 4 wk after exposure, and the fiber numbers an...

The effect of asbestos and stone-wool fibres on some chemokines and redox system of pulmonary alveolar macrophages and pneumocytes type II.

The in vitro effect of stone-wool has been studied in primary cultures of pulmonary alveolar macrophages (AM) and type II pneumocytes (T2) by morphological, biochemical and immunological methods. UICC crocidolite was applied as a positive control. Although stone-wool brought about frustrated phagocytosis, it did not induce serious membrane damage, whereas crocidolite gave rise to very severe me...

Surfactant protein A enhances the phagocytosis of C1q-coated particles by alveolar macrophages.

Surfactant protein-A (SP-A) plays multiple roles in pulmonary host defense, including stimulating bacterial phagocytosis by innate immune cells. Previously, SP-A was shown to interact with complement protein C1q. Our goal was to further characterize this interaction and elucidate its functional consequences. Radiolabeled SP-A bound solid-phase C1q but not other complement proteins tested. The l...

CHROMOSOMAL PROTEINS IN PULMONARY ALVEOLAR MACROPHAGES

In this study the nature of chromosomal proteins, his tones and nonhistone in resident alveolar macrophages was investigated in comparison to peritoneal neutrophils and calf thymus proteins. Cells were obtained by lavaging the lung and after purity determination they were subjected to fractional extraction procedures. Proteins were then analysed on SDS polyacrylamide gels and densitometric...
